Abstract:Acid invertase was isolated from Sorghum bicolor (L.) Moench. Activity was determined in a crude enzyme preparation by determining the increase in glucose and fructose and the decrease in sucrose. TMS derivatives of the sugars were analyzed using gas-liquid chromatography. A mean decrease in sucrose of 30.6±4.5% accompanied by an increase in glucose and fructose was observed after 4 hr. After 2 hr the percent inversion ranged from 7%-24% indicating variable concentrations of active enzyme in the different enzyme preparations. A nonstoichiometric increase in monosaccharides was observed. In 3 experiments glucose increased 20.6±0.6%. Fructose increased 9.6±4.5% indicating the presence of an isomerase. The mg of soluble sugar per g of root tissue ranged from 1.0-5.6 mg indicating large differences in the physiological status of the tissue used in different enzyme preparations.
