Abstract:ABSTRACT: Sinclair miniature swine are a useful animal model for studying malignant melanoma. As this tumor has been suggested to be sex steroid sensitive, it would be useful to produce a steroid free or reduced steroid condition in this animal model. The adrenal cortex produces significant amounts of steroid hormones and this production must be suppressed to establish this hyposteroidal condition. O,p'-DDD, a supposed specific adrenocorticolytic drug, was used for this purpose in this study. O,p'-DDD inhibited basal cortisol levels following a 100 and 1000mg/kg/day treatment regimen. The 500 mg/kg/day dose did not significantly alter basal cortisol levels. Cortisol response was inhibited in all treatment groups but not in a dose related manner. Furthermore, although only testosterone response had a statistically significant reduction at 500 mg/kg/day suggesting both a treatment and dose effect, basal testosterone levels were essentially unchanged. Aldosterone basal and response levels were inhibited albeit not to a statistically significant degree. Adrenocortical and hepatotoxicity were observed at all treatment levels, and neurotoxicity was implicated in the deaths of two of the five animals lost during the study. The lack of a dose effect by o,p'-DDD may have been due to maximal gastrointestinal uptake or saturation of specific intracellular o,p'-DDD binding sites.
