Abstract:ABSTRACT: Periostin is an extracellular matrix protein that binds the [alpha]v[beta]₃ and [alpha]v[beta]₅ integrins, and activates focal adhesion kinase (FAK) and Akt. Periostin plays a significant role in tumor growth by promoting angiogenesis and preventing apoptosis during cancer progression. Therefore, we hypothesized that periostin loss would have a beneficial effect on survival and prevent tumor growth in colorectal cancer. To test our hypothesis, we investigated the effect periostin loss had on tumor growth and survival in Apc[min/+] mice, a mouse model for colorectal cancer. Our studies demonstrate that loss of periostin in Apc[min/+] mice results in a marked decrease in the number of intestinal polyps and an increase in animal survival. Additionally, the periostin loss results in a decrease in cell proliferation and an increase in apoptosis. These data indicate that periostin promotes tumor formation and progession, and is a potential drug target for clinical treatments of cancer.
