| Summary: | Six positional candidates genes were evaluated for their effects on previously identified meat tenderness QTL on BTA5. Candidate genes were selected using a human-bovine comparative map. Four genes, MMP19, WIF1, WNT1, WNT10B, and two genes, MYF5 and MYF6, were selected as candidate genes for WBSF and OTEND QTL, respectively. BACs were isolated for all candidate genes with the exception of WNT1. Using FISH, four candidate genes (MMP19, MYF5, MYF6, and WIF1) mapped to BTA5 and one paralog gene (MYF4) mapped to BTA16. RH mapping led to three new gene assignments on the BTA5 RH map (MMP19, WIF1, and WNT10B) while confirming one previously known position (MYF5). A polymorphic microsatellite (16R4C12.3L12) was developed using sequence isolated from BAC clones containing candidate genes MYF5 and MYF6 enabling their first time placement on the bovine linkage map. This microsatellite mapped to a QTL for OTEND in half-sib families segregating for the QTL indicating MYF5 and MYF6 are strong candidates for OTEND QTL on BTA5. Sire QTL effects were significant with homozygous individuals for the Angus allele being 2.8 OTEND units less than homozygous individuals with the Brahman allele. MYF4 was mapped to an OTEND QTL on BTA16 giving further support to a relationship between the MYF family of genes and OTEND. While MMP19, WIF1, WNT1 and WNT10B failed to map to the QTL for WBSF, consensus map positions for these genes helps improve the resolution of the bovine genomic map for BTA5 as well as providing information for future studies. Using novel two-point diagnostic plots allowed for easy visual analysis of the RH map. Discordant plot patterns in the interval between zero and four rays indicate a block inversion in this region of the BTA5 RH map order. Breakage probabilities when plotted against distance in Rays, plotted below the estimated values indicating map inflation. Gene expression analysis results have been inconclusive up to this point. Preliminary data using Real-Time PCR indicated gene expression of MYF6 between esophagus, heart and neck muscle tissue collected shortly after slaughter. Fluorimetry and densitometry results failed to confirm that differential gene expression of MYF6 is taking place. |
