| Summary: | In order to determine whether hindlimb unloading (HU) alters the extracellular matrix of skeletal muscle, male Sprague-Dawley rats were subjected to 0 (n=11), 1 (n=11), 14 (n=13), or 28 (n=11) days of unloading. Remodeling of the soleus and plantaris muscles was examined for collagen abundance biochemically via measurement of hydroxyproline (OH-Pro) and estimated histologically with picrosirius red (PSR) staining. The cross-sectional areas of type l and III collagen were also determined with the PSR staining. Total hydroxyproline concentration was not different in the soleus or plantaris muscle with HU. Following 14 days of HU, however, the relative proportion of type l collagen in the soleus muscle (52±7%) decreased relative to control soleus muscle (68±5%). Correspondingly, there was an increase in the proportion of type III collagen in soleus muscle with 14 days of HU (control 33±5%; l4dHU 48±7%). A similar shift in collagen isoform was also determined with 28 days of HU. In addition, no changes in the proportion of type I and III collagen occured in plantaris muscle with HU. No alterations were evident in type l or III collagen mRNA in soleus muscle following HU. The data demonstrate that HU induces a shift in the relative proportion of collagen isoform (type I to III), which occurs concomitantly with a slow- to fast-twitch myofiber transformation in rat antigravity muscle. |
