Summary:In vitro, in vivo and in ovo studies were designed to investigate the effects of monoclonal antibodies (MAbs) raised against embyonic chicken adipocyte plasma membranes, on the development of adipocytes and adipose tissue in chickens. Adipocyte plasma membranes isolated from 18-day chicken embryos were used as the antigens to immunize BALB/c mice. Antibody-secreting hybridomas were produced by fusion of immunized mouse lymphocytes with SP 2/0 myeloma cells. Monoclonal antibody immunofluorescence assay of primary cell cultures from embryonic tissues demonstrated specific binding to cells containing intracellular lipid droplets. For in vitro investigation, 4 individual MAbs with complement significantly reduced (P<0.05) the number of fat cell clusters that developed in chicken adipocyte primary culture by 40-60%. These effective MAbs in presence of complement also reduced (P<0.05) mean cell width, and apparent cell area or cell cluster area of lipid-containing cells. Neither MAb nor complement alone reduced fat cell cluster number, cell size, or cluster size. Treatment with pools of 2, 3 and 4 MAbs decreased the total amount of MAb protein concentrations required to effectively reduce fat cell cluster number. For in vivo investigation, MAbs were injected into the right pectoral adipose depots of l4-day and newly hatched chickens. Local in vivo Mab injection significantly reduced (P<0.05) the injected fat pad weight without affecting other major organs and non-injected fat pad weights for short-term (2 weeks) and long-term (6 weeks) studies. Total body weight and feed intake per bird were not affected by the treatment. For in ovo investigation, MAbs were injected intravenouly into the allantoic circulation of fertile eggs at 15-day of embryogenesis. Adipose tissue, major organ and total body weights at 14 days of age were not affected. By 42 days of age, abdominal fat pad mass was significantly reduced (P<0.05) by MAb injection, while total body weight and feed intake were not affected Monoclonal antibodies were developed that were specific for lipid-containing cells. These antibodies in the presence of complement reduced adipocyte development in vitro and reduced adipose tissue mass in vivo and in ovo.
