Summary:Studies on the classification of bacteria using Fourier transform infrared spectroscopy were completed. Nine different species of bacteria were selected for this study. Different methods for growing the bacteria were investigated. This study evaluated various sample preparation techniques and a selected polyethylene film method. In this procedure the bacteria are spread on the film and vacuum desiccated for thirty minutes. Then the infrared spectra of the bacteria were collected. Since neither the concentration of bacteria nor the chemical composition is known, the traditional classification methods are not suitable. Classification methods such as cartesian distance and analysis of variance that require prior normalization were applied to the spectra and classification results were tabulated. Success rates for classification using cartesian distance metric was 73.33% to 13.33%. Classification by direction cosines and correlation does not require prior normalization. Direction cosines use the angle between the unknown spectrum and the reference spectrum to classify. Among all the methods this method yielded the best results. Error analysis for direction cosines shows that 86.67% to 33.33% of bacteria were successfully classified. Except for E. coli DH5A and E. coli BL21 all the other bacteria of different species were classified at a rate greater than 73%.
