Summary:Ancient DNA extracted from Pecos River genre rock paintings from the Lower Pecos River region of Texas was reported in 1996. That work indicated that the organic binder/vehicle source material was obtained from a member of the Order Artiodactyla. My research attempted to replicate that earlier work and to obtain a species level determination of the organic binder/vehicle source material. The mitochondrion gene Cytochrome C Oxidase Subunit II (COII) was selected because of the large slumber of copies of the mitochondrion genome available relative to the nuclear genome, and the gene has sufficient sequence variation to obtain a species determination for any extracted DNA. Unlike the previous study, a new laboratory where no mammals had been studied was available. However, after multiple extraction attempts that used three different extraction protocols, no ancient DNA was found. Unless independently confirmed elsewhere, the prior report of ancient DNA in Pecos River genre rock paintings is invalid.
