| Summary: | Recent reports of endocrine disruption in wildlife and human populations have implicated organochlorine compounds as agents responsible for various reproductive and developmental abnormalities. In particular, organochlorine compounds such as PCBs and pesticides have been characterized as environmental estrogens. Several studies have shown synergistic estrogenic interactions between several organochlorine compounds in in vitro assays, suggesting that the estrogenic potency of organochlorine mixtures may be underestimated. Studies presented herein show that several organochlorine compounds have minimal estrogenic activity and significant antiestrogenic activity in in vivo assays. Additionally, previously reported synergistic interactions between organochlorine compounds was not observed in multiple in vivo and in vitro assays. Studies presented in this thesis have also identified a new class of naturally-derived aryl hydrocarbon receptor-based antiestrogens/antitumorigens that exhibit low toxicity and may be useful for clinical treatment of breast cancer. These studies indicate that the estrogenic activity of organochlorines may be overestimated and that human exposure to antiestrogens may be more prevalent. |
