| Summary: | Seed storage protein gene expression is under strict developmental control. Seed storage proteins are expressed in a spatial, temporal and quantitative manner. Sunflower (Helianthus annuus) seeds accumulate predominately two classes of storage proteins, the 11S globulins and 2S albumins. A series of 5' deletions was employed to identify functional regions of the 2S albumin gene, HaG5 , and the distal promoter region of the 1 IS globulin gene promoter, HaG3A. The -252 to +29 region (PPR) of HaG5 conferred seed-specific expression in developing Arabidopsis embryos, the -252 to -452 region (DPR) contributed to quantitative expression. The distal promoter regions of the 11S globulin, helianthinin, conferred variable expression in developing tobacco embryos suggesting the presence of positive and negative elements. Gene regulation is mediated through the interactions of sequence-specific DNA binding proteins. The PPR and DPR of HaG5 interact with similar sunflower seed-specific nuclear proteins. Sunflower nuclear proteins bind putative cis-elements of the 2S albumin PPR, these include an E-box motif which is conserved among numerous 2S albumin genes. |
