| Summary: | Although the basic structure of the ovary of domestic hens has been described, the factors controlling maintenance of the follicular hierarchy have yet to be completely defined. Luteinizing hormone (LH) and follicle stimulating hormone (FSH) have been the most investigated molecules with regard to ovarian follicle steroidogenesis, and to a lesser extent follicular growth. In the present manuscript, we report effects of FSH or LH on in vitro []H-Thymidine ([]H-TdR) incorporation, indicative of cellular DNA synthesis, by granulose cells from follicles of selected maturity, F1 , F3, or F5-6, in the ovarian hierarchy. These investigations have shown ovine. or chicken-derived FSH to be capable of stimulating DNA synthesis in granulose cells from all follicles collected. Stimulatory effects of FSH on granulose cell DNA synthesis were dependent on the position of the follicle in the hierarchy. FSH was most efficacious in stimulation of granulose cell DNA synthesis in granulose cells from the F1 or F5-6 follicles with lesser effects on granulose cells from the F3 follicle. The effects of ovine-derived (oLH) or chicken-derived LH (cLH) on in vitro DNA synthesis were dependent upon the specific stage of follicular maturity. Factors other than pituitary gonadotropins have been reported to affect hen ovarian follicular function. Previously, our laboratory reposed isolation and partial characterization of bursal anti-steroidogenic peptide (BASP) from the chicken bursa of Fabricius. This molecule has potent inhibitory activities on chicken ovarian granulose cell steroid biosynthesis and mitogen-induced DNA synthesis in neonatal chick bursal lymphocytes and peripheral blood lymphocytes derived from chicken, rat, or human sources. In the present experiments, BASP significantly increased DNA synthesis in granulose cells from all follicles collected. Again, variable effects of LH on granulose cell DNA synthesis, either alone or in combination with BASP, were observed. In combination with FSH, BASP resulted in additive or synergistic increases in DNA synthesis. Experiments in the present manuscript indicate that FSH and BASP, and to a lesser extent LH, modulate granulose cell DNA synthesis in vitro. Furthermore, these data suggest a maturation-dependent differential response of granulose cells to these agents. |
