Summary:The homozygous leaner mouse exhibits the most severe ataxia among the approximately thirty currently known cerebellar mutant mice. Morphologic studies have revealed neuronal degeneration in the cerebellar cortex of the leaner mouse. The genetic etiology of the leaner mutation has recently been discovered to be a splice-site mutation in the voltage-sensitive calcium channel subunit α₁[] gene resulting in novel α₁[] protein(s). The aims of this dissertation were to investigate the molecular and cellular changes induced by the mutation in the α₁[] gene of the leaner mouse. The involvement of apostasis in neuronal degeneration in the leaner cerebellum was examined by the techniques of in situ end-labeling of DNA strand breaks and electron microscopy. The expression of the α₁[] mRNA and protein in the leaner cerebella was investigated with in situ hybridization histochemistry and immunocytochemical techniques, respectively. Intercellular free calcium level of leaner cerebellar neurons was determined by fura-2 acetoxymethyl calcium fluorometric technique. Finally, differential display technique was used to isolate genes whose expression was altered in the leaner cerebellum.
