The research described herein examines the MALDI ion research described herein examines the MALDI ion observed mass resolution and describes how these limitations can be overcome to yield high resolution MALDI-TOF mass spectra. The instrumental requirements for acquiring high resolution MALDI-TOF mass spectra are then discussed in detail, including the advantages of high resolution mass spectra. One clear advantage of high resolution mass spectra is that it allows the possibility to perform accurate mass measurements. The utility of MALDI-TOF accurate mass measurements in the analysis of biomolecules is discussed herein and focuses on the identification and characterization of proteins using accurate mass MALDI-TOF of enzymatically generated peptide maps. |
