| Summary: | To positionally clone the polled/horned locus (POLL) phics. in cattle, genetic and physical fine maps around the POLL locus were constructed using both published and newly-developed markers in a reciprocal backcross and F2 Angus x Brahman beef cattle population. Ten microsatellite markers, 162M1, AGLAI7, 8M6438, WRA212, SODIMICR02, BMS 1928, TGLM9, ARO24, ARO9 and WRAI 17 were used to develop a genetic fine map around POLL with an average marker interval of 0.87 cM. Among these markers, 162M1 and SODIMI/ROZ were developed in our lab in order to increase the number of informative meioses for the markers AGLAI7 and SODI . The order and distance among these markers in the genetic fine map is 162M1-(0.0 cM)-AGLA17-(1.6 cM)-BM6438-(0.0 cM)- J'OfZ-(1.3 cM)-1NRA212-(0.8 cM)- SOD1MlCRO2-(0.7 cM)- TGLA49-(0.0 cM)-ARO24-(1.1 cM)-ARO9-(2.3 cM)- BMS 1928-(0.9 CMI-INRAI 17. To confirm the location and ordering of these markers, physical mapping using FISH was done for nine markers, including SODI, SST, AGLAI7, 8M6438, IZAR, BMS 1928, TGLA49, WRAI 17 and KAP8. The order obtained from both the linkage analysis and FISH experiment was the same for the seven common markers, SODI, AGLAI7, 8M6438, BMS1928, TGLA49, INRAII; and KAP8. With these fine maps in hand, we can begin to pursue the positional cloning of the POLL gene. |
