Summary:The sequence analysis of tomato cDNA c34, a candidate gene
for the jointless gene from tomato, revealed that the deduced
amino acid sequence encoded 506 amino acid residues and
contained two putative activation domains of transcription
factors: one serine/threonine rich region and one acidic
region. The gene family of c34 is present in distantly
related species, including Arabidopsis thaliana, Nicotiana
tabacum, Oryza sativa, and Pisum sativum. The sense c34
construct failed to complement the jointless mutant.
Surprisingly, a transient epinasty growth phenotype was
observed in young tomato in both sense and antisense c34-
transformed young wild-type tomato plants. In c34-
transformed tobacco, overproduction of the c34 transcript
caused pleiotropic alterations in the mature tobacco plants,
which include reduced plant height, decreased elongation of
peduncle and corolla, and suppressed pigmentation of petals.
In c34-transformed Arabidopsis, an incidence of a gene
silencing effect between two homologous genes was observed.
By impairing the host gene ATTS2203 with its potential tomato
homolog c34 in Arabidopsis, the sense and antisense c34-
transformed Arabidopsis exhibited conspicuous alterations in
the inflorescence structure such that the spiral phyllotaxy
was converted to the whorled phyllotaxy, and the
inflorescence growth pattern was switched from indeterminate
to determinate by producing terminal flowers at the
inflorescence apex. Based on the evidence of sequence
analyses and
transgenic assays of tomato, tobacco, and Arabidopsis, I
infer that c34 might function as a transcription factor
involved in controlling hormone perception, translocation, or
an appropriate hormone ratio. In Arabidopsis, the c34
homologue plays a notable role in the early stages of flower
development.
