Summary:The gene clusters and introns in chloroplast DNAs of
charophytes, the closest green algal group to land plants,
are poorly known. Here, the genes of four operons unique to
land plants (rpl23, clpP, psaA, and psbB operons) were
investigated in Spirogyra cpDNA, and psbB operon genes were
also investigated in Coleochaete and Chara cpDNAs. In
addition, the presence/absence of introns in 6 genes (petB,
petD, rpl2, rpl16, clpP, and ndhA) was determined in
Spirogyra. Spirogyra contains three gene clusters (rpl23,
clpP, and psaA) and 6 introns in 5 genes (petD, rpl2, rpl16,
clpP, and ndhA), which are shared by land plants. These
results support the placement of Spirogyra, along with other
charophytes, close to the land plant lineage. However,
unlike land plants, Spirogyra has the psbB operon genes in
two separate gene clusters, and petD contains an intron but
petB does not contain an intron. Unlike Spirogyra,
Coleochaete and Chara cpDNAs contain a psbB gene cluster
similar to land plants. Coleochaete contains introns in both
petB and petD, and Chara has an unusually large petB intron
but no intron in petD. The detailed characterization of the
psbB gene cluster, its intron distribution, and sequence
analysis using psbB, petB and petD gene sequences indicate
that Coleochaete is most closely related to land plants, and
that Spirogyra is basal to Chara, Coleochaete, and land
plants.
