| Summary: | Glucagon-like peptide-1 (7-36) amide (GLP-1), a glucose- dependent insulin secretagogue, is released by the distal ileum in response to food entering the gut. Infusion of GLP- 1 into the brain ventricles dose-dependently inhibits feeding. The intent of the present experiments was to determine whether infusion of GLP-1 into the hypothalamic paraventricular nucleus (PVN) would suppress feeding through the induction of satiety, and to determine whether the PVN serves a primary role in the feeding-inhibitory action of central GLP-1. Experiment 1 revealed that PVN infusion of 100 and 200 ng GLP-1 dose-dependently reduced consumption of a palatable liquid diet. Experiment 2 demonstrated that PVN infusion of GLP-1 (100 and 200 ng) did not induce malaise as assessed by a conditioned taste aversion paradigm, and Experiment 3 demonstrated that PVN infusion of GLP-1 (100 and 200 ng) did not significantly alter locomotion. Experiment 4 revealed that PVN infusion of 100 and 200 ng GLP-1 significantly reduced pellet chow and water intake, but that infusion of GLP-1 (100 and 200 ng) into brain sites ventral and/or lateral to the PVN did not suppress feeding or drinking. Experiment 5 demonstrated that PVN infusion of GLP-1 (100 and 200 ng) reduced feeding without disrupting the behavioral satiety sequence, suggesting that exogenous GLP-1 within the PVN promotes satiety. Finally, Experiment 6 revealed that the anorexia induced by lateral ventricular infusion of GLP-1 was abolished by bilateral electrolytic lesion of the PVN. These results suggest that the PVN mediates the anorexia induced by ventricular infusion of GLP- 1, and that exogenous GLP-1 within the PVN promotes satiety. |
