| Summary: | Neural tube defects (NTDS) persist as one of the most common congenital malformations in the United States, affecting approximately 1 out of every 1000 liveborn infants. While it is known that both genetic and environmental factors contribute to the pathogenesis of these defects, the specific genes involved in conferring susceptibility to heat-induced NTDs have not been identified. In order to identify these genes, this study utilized two well-characterized murine models for heat-induced exencephaly, in combination with a genetic screening approach, differential display, to characterize differential embryonic gene expression within the neural tube of these highly inbred strains at gestational day 8.5. As a result of this approach, this study reports the identification of two candidate genes., growth arrest specific gene 5 (gas 5) and the peroxiredoxin AOE37-2, for the NTD susceptibility observed in these strains. The role of each gene and the implications of these findings will be discussed in this work. By characterizing candidate genes for NTD susceptibility in a mouse model, it is our goal to ultimately provide improved prenatal screening for these defects, as well as to aid in their future prevention. |
