Summary:Comparative gene mapping may be accomplished by a variety of
methods including synteny mapping and linkage mapping. The
former is generally achieved by somatic cell mapping of
homologous type I loci in different species to identify
evolutionarily conserved syntenic segments, while the latter
addresses gene order and linkage conservation within
conserved syntenic segments. Somatic cell mapping provides
little if any information about the order of loci within a
conserved segment. A study of conservation of gene order
within conserved segments is necessary to generate refined
comparative maps capable of providing information on internal
rearrangements in chromosome evolution. This dissertation is
a study of the evolutionary relationship of human chromosome
17 (HSA17) and bovine chromosome 19 (BTA19) both in terms of
synteny conservation and internal structural rearrangement.
First, eight comparative anchor loci on HSA17 (CHRNB1, CRYB1,
MYL4, MPO, NF1, P4HB, THRA1, and TP53) were assigned to BTA19
using cattle X rodent somatic hybrid cell lines. This
completed the bovine synteny mapping of human chromosome 17
comparative anchor loci, all of which have been
mapped to bovine chromosome 19 and mouse chromosome 11
(MMU11). Second, conservation of gene order within the
conserved segment represented by HSA17, BTA19, and MMUll was
explored by different strategies including linkage studies in
an international bovine reference family (IBRF) panel and in
a bovine interspecific hybrid backcross (IHB) panel, and by
whole genome radiation hybrid (WG-RH) mapping. Eight
comparative anchor loci on HSA17 and five microsatellite
markers were included in the BTA19 linkage map by the IBH
method. Seven HSA17 comparative anchor loci and nine
microsatellite markers were placed on the WG-RH map of BTA19.
The orders obtained by the three mapping methods are in
agreement. This study expands our knowledge of physical,
linkage and comparative maps of BTA19 and introduces two new
strategies, IHB and WG-RH mapping for cattle gene mapping.
These new methods have the advantage of being more efficient
than traditional bovine linkage studies for mapping type I
loci and their successful application in cattle will greatly
benefit the comparative mapping of the cattle genome.
