| Summary: | A safe and repeatable non-surgical embryo collection technique was developed for the chimpanzee. The embryo recovery rate was 28% (n = II 3) for all collection attempts, and as high as 50% ( n = 12) for individual chimpanzees with known fertility. Medium return averaged 92%. Embryo recovery rate was highest when ovulation timing was estimated using the combination of the urine LH peak and the perineal detumescence score. Male chimpanzees were evaluated behaviorally, prior to training for semen collection with the artificial vagina (AV). Behaviors predictive of their acceptance of the AV were identified and used to rate prospective candidates for training. The mean volume of chimpanzee semen collected ( n = 25) was 0.99 ml, concentration was 479 X 10' sperm/ml, motility was 57%, normal morphology was 82%, and rate of forward movement (RFM) was 5. Chimpanzee semen frozen with Test Yolk Citrate extender was thawed and used for artificial insemination, resulting in the recovery of 1 pronuclear stage embryo. Semen collections were attempted on 26 gorillas by rectal probe electroejaculation (RPE). The mean volume recovered (n = 22) was 0.8 n-J, motility was 22%, concentration was 140 X 106 sperm/ml, normal morphology was 48%, and RFM was 3.5. The mean testicular measurements ( n = 24) were 42.5 X 26 mm. Calculated testicular volume was highly correlated with body weight ( r = 0. 8144), but not with total concentration nor motility. Washed, cooled gorilla sperm maintained motility for up to 46 hours in Ham's F-10 medium. Records from a human infertility clinic were analyzed to investigate the feasibility of applying techniques developed for dealing with male factor infertility in humans to the gorilla. At least one category of male factor infertility was noted in 87% of the human cases presented for IVF ( n = 215), yet fertilization rates were similar to cases fertilized with sperm from males with normal sperm factors ( 71% and 76%, respectively). Parameters of fresh semen were not highly correlated with IVF fertilization rates with processed semen, indicating that pre-treatment of sperm used for IVF may overcome some of the effects of male factor infertility. |
