Summary:Fifteen mononucleotide and dinucleotide repeat microsatellite markers were isolated from a bovine library and characterized- Seven of these markers, in addition to 96 other published markers were used to develop < 20 cM sex-averaged and sexspecific genetic maps for chromosomes I through 6, 21 and 23, which spanned 740.9 cM or approximately 27% of the 2,800 cM bovine genome, when scored in a reciprocal backcross and F2Angus (AA) x Brahman (BB) beef cattle population. An unassigned linkage group, comprising cattle population. An unassigned linkage group, comprising microsatellites OarFCB II, BM2113, BMI223 and BM6444 was integrated into the chromosome 2 map. The degree of saturation of these eight chromosome maps was sufficient to identify economic trait loci (ETL) having a major effect on several growth and carcass traits. A single locus multiple regression model based on interval mapping was used to identify ETL with LOD > 2.0 in 209 slaughter progeny. ETL reported are followed in parenthesis by chromosome, 12a I /op value denoting the distance between alternate homozygotes in standard deviation units, and homozygote performance at the estimated ETL position. These include: weaning weight (2, 0.91, AA > BB), average daily gain from birth to weaning (2, 0.92, AA > BB), out of Angleton weight - entry to feedlot (2, 0-79, AA > BB), out of McGregor feedlot weight (2, 0.84, AA > BB), slaughter weight, (2, 0.89, AA > BB), hot carcass weight (2, 0.89, AA > BB), dressing percentage (5, 0.94, AA > BB and 23, 0.84, AA < BB), rib eye area, 2, 0.86, AA > BB- 5, 1.1, AA > BB- 6, 0.91, AA < BB and 21, 0.80, AA > BB), kidney, pelvic and heart fat (2, 0.95, AA < BB and 5, 0.917 AA > BB), calpastatin, (5, 0.82, AA > BB), cholesterol (1, 1.08, AA > BB) and stearic acid (4, 0.95, AA < BB).These results provide the initial localization of ETL for the application of MAS in future beef cattle breeding programs. Utilization of comparative mapping information and a Bacterial Artificial Chromosome library developed in this laboratory will enable the positional cloning of the most economically important ETL.
