Summary:A panel of 40 Bos taurus x Bos gaurus (gaur) interspecific
hybrid backcross offspring was constructed to be used as a
tool for refining and improving the resolution of the bovine
gene map. of the bovine gene map. The utility of
interspecific backcrosses has already been proven in the
development of the mouse gene map, as a result of increased
probability of parental heterozygosity at a given marker.
The gaur (2n=58) is characterized karyotypically is
characterized karyotypically by a 2;28 Robertsonian
translocation with respect to the cattle karyotype (2n=60).
This translocation was used as a centromeric marker to
directionally orient linkage maps of Bos taurus chromosomes
BTA 2 and BTA 2 8. Markers tested in the linkage analysis
represented genes or DNA segments previously placed on bovine
maps, as well as markers chosen from bovine-humanmouse
comparative maps. Additionally, fibroblast cultures from
fetal tissue of each offspring were analyzed cytogenetically
at early passages. In all cases, the translocation
chromosome was inherited in a balanced
condition, and only one cell line possessed a chromosome
abnormality. The trisomic line was karyotyped as 61,XXX.
Among the DNA markers analyzed, MYF6, SKMC, COX4, COX6AH,
COX6AL, COX8H, COX8L, ALPI, CHAT, DBI, and SRM represent
additions to the bovine gene map.
