| Summary: | The bovine genomic map was expanded with the primary goal of comparative gene mapping. Multiple approaches were used, including the synteny mapping of three multigene families. Five members of the G protein gene family, GNAS, GNAI, GNAT, GNBT and GNGT were assigned to bovine chromosomes 13, 22, 22, 16 and 19, respectively, using either PCR or Southern blot analysis of DNA from bovine x rodent hybrid somatic cell lines. Members of the casein kinase 11 gene family, CSNK2Al and CSNK2A2 were mapped in the same hybrid cell panel to bovine chromosome 13 and 5, respectively. Using the same strategy, endothelin receptor genes, ETAR, ETBR and four other bovine genes, MCP1, CALLL, PI and PFKFBL were assigned to bovine chromosome 17, 12, 19, 3, 21 and X. Four gene loci, ETBR, MCP1, CALL L and PFKFBL revealed variation by RFLP analysis. Six genes designated as comparative anchor loci on HSAL 9, were mapped in cattle. Four genes, CALR, EPOR, JUND and JUNB located on the short arm of HSA19, were assigned to bovine chromosome 7, whereas the other two, CEA and PRKCG, located on the long arm of HSA19, were assigned to bovine chromosome 18. Polymorphic microsatellite markers are important for the development of the bovine linkage map. Cloning and screening microsatellites from a sizeselected library were attempted, resulting in isolation of one polymorphic microsatellite, designated TXQG1. Both physical and genetic analysis assign TXQG 1 to bovine chromosome 1 0. Using a Bos taurus x Bos gaurus interspecific hybrid backcross panel, a genetic linkage map of bovine chromosome 7 was established, including five Type I loci and six Type 11 loci. The five Type I loci were also incorporated into the current linkage map of bovine chromosome 7 by analyzing the international reference family. Comparison of the two linkage maps resulting from different cross strategies revealed a consistent gene order, suggesting the usefulness of this interspecific cross panel in establishing the primary bovine linkage map. |
