| Abstract: | The goals of the bovine gene mapping effort are to develop a comparative physical map and a linkage map with highly informative markers evenly spaced every 25 centimorgans (cM). The comparative map gives insights into the evolutionary rearrangements that have occurred between the bovine and other species. The linkage map allows the correlation of physiological traits with given genotype(s) at one or more loci. There is an imperfect linear relationship between the assignment of loci on the comparative and linkage maps due to differences in recombinational frequencies between sexes and chromosomal locations. The physical map of a species is a linear arrangement of loci on a chromosome derived from methods other than genetic recombination. The comparative physical map is a composite of physical maps of homologous loci of two or more species. The comparative physical map consists of type one loci (coding genes conserved among species) while the linkage map consists of primarily type two loci (noncoding genes that are species-specific and highly polymorphic). Type one loci are often excluded from the linkage map due to lack of polymorphisms, leading to the development of two independent maps. The purpose of this project was to assign a total of 5 type one and 11 type two loci to the bovine physical map in an effort to provide loci that could be used for both maps. Single strand conformation polymorphisms (SSCFs) provide a means of fusing the two maps by detecting variations in assigned type one loci that are subsequently used in the linkage map. SSCPs were used to aid in the detection of mutations in 12 of 15 loci, 11 of which were type one loci. Linkage analysis was then conducted for ARVP, OXT, DU11S1, and PRNP for U11 and for BSPN and PLANHI. Although no linkage was detected, the use of SSCPs has proven to be a useful tool in the anchoring of the bovine linkage and physical maps. |
