| Abstract: | The recognition of and selection for outstanding genetic material has been crucial to the successful improvement of desirable traits in cattle. The advent of molecular biology has made it possible to identify the causes of quantitative phenotypic variation between individuals. Localizing the quantitative loci responsible for production, health, and efficiency traits is a high priority for the continued improvement of cattle. Two approaches are currently used to identify quantitative trait loci (QTLs). The implications of the physiological effect of certain loci can make them candidates for the study of quantitative traits. Alternatively, a genetic map can be used as a mechanism to scan the genome for QTLs. This study represents preliminary work in the search for QTLs. A group of candidate loci, the prolactin related proteins and placental lactogen, were studied for their genomic distribution and potential for QTL analysis. This gene family was found to map to bovine syntenic group U20. All members show natural variation in cattle populations and initial studies indicate a close physical relationship among these loci. The development of polymorphic markers will be important for the development of the bovine linkage map. A size selected bovine library was generated and screened for the presence of microsatellites. The library constructed was found to be useful for the cloning of microsatellites other than TG repeats which are present in a larger bovine repeat inherent in the library. The development of a bovine genetic map was investigated using a novel approach for the mapping of loci in cattle. The map generated from this work is based on a sequence tagged site approach. The sequence tagged site map consists of small regions of sequenced DNA recoverable through amplification by the polymerase chain reaction. Both type I loci (conserved protein coding genes) and type II loci (anonymous polymorphic markers) were mapped and make up the new sequence tagged site map. A total of ten type I loci and twenty-four type II loci were assigned to syntenic groups. Eighteen percent of the bovine map now consists of sequence tagged sites. |
