Abstract:We characterized ventricular/vascular coupling by measuring changes in systemic hydraulic power in young and adolescent Yorkshire-cross pigs fed a normal pig diet (n=9, Cl), adolescent Sinclair-cross (miniature) pigs fed a normal pig diet (n=5, C2), old Hanford-cross pigs fed a normal diet (n=3, 0) and two groups of hyperlipidemic diet fed Yorkshire-cross (n=4, H1) and Hanford cross (n=6, H2) pigs . Left ventricular pressure, aortic root pressure (AP) and aortic root flow (Q) were measured at baseline and during transient changes in AP. Steady potential power (E[p,s]) , oscillatory potential power (E[p,0]) and the oscillatory potential to total potential power ratio (E[p,0]/E[p.T]) were calculated. Cl, O and H1 pigs were given intraarterial injections (IA) of Angiotensin II (All) (.01 mg/kg), and Nitroglycerin (NG) (.1 mg/kg). C2 and H2 pigs were given Acetylcholine (ACh) (10μg/kg) and 5- Hydroxytryptamine (5-HT) (10 μg/kg). All measurements were made prior to recirculation of the vasoactive agents. Baseline conditions were not significantly different within Baseline conditions were not significantly different within diet groups. HI and H2 exhibited higher AP, lower Q and lower EP, S at baseline compared with Cl and C2. C1 exhibited appropriate changes in the parameters following the injections. The old pigs exhibited similar changes in AP and E[p,s] following All and NG, but the magnitude of the AP response following AII and NG was significantly blunted compared with C1. E[p,s] was significantly decreased for the O pigs following NG compared with C1. E[p,0]/E[p,T] following All and NG were not significantly different between C1 and O. H1 exhibited a decrease in E[p,T] following All and increases in E[p,T] following NG. H2 pigs exhibited an augmented reponse to ACh although not significantly different compared with C2. No differences were observed following 5-HT between C2 and H2 pigs. Evaluation of systemic hydraulic power was used to determine if age or diet would alter the total and pulsatile load. Alteration in these parameters due to age or hyperlipidemia were observed for both endothelium-dependent and independent vasoactive agents.
