Abstract:The inner-membrane pools of F-pilin in Escherichia coli K-12 strains containing the conjugative F plasmid were assayed to determine the acetylation state of the pilin subunits. Monoclonal antibodies directed against either the acetylated N-terminus or against an internal epitope of the pilin molecules were used in Western blot analyses. Flac tra amber strains, insertion mutant strains, Hfr deletion strains and strains containing portions of the F transfer region on plasmids were all used to determine the location of the F gene required for acetylation of the F pilin subunit. A locus which is required for this modification was found to be located at the extreme promoter-distal end of the F transfer region in a previously uncharacterized locus named traX.
