| Abstract: | Suitable electrophoretic techniques were developed to detect activities of thirteen cotton isozyme systems using horizontal starch gel, vertical starch gel and polyacrylamide discontinuous tube gel systems. Peroxidase isozymes of cotton were also detected after iso-electric focusing on a mini gel system.Interspecific chromosome substitution stocks and crosses between chromatin-deficient stocks of TM1(Gossypium hirsutum) and PIMA 3-79 (G. barbadense) were utilized to locate positions in the chromosomes of three biochemical markers, Pgm-1, Pgm-2, and Aco-1. The Pgm-1 locus was located to the short arm of chromosome 22 of G. hirsutum. The Pgm-2 locus was located to the long arm of chromosome 12 of G. hirsutum at a distance of 58 (plus or minus sign) 0.07 cM from the centromere. Aco-1 was found to be not located in chromosomes 1, 2, 3, 4, 6, 7, 10, 12, 16, 17, 18, 22 or 25.Genetic diversity within the germplasm of Upland cotton (G. hirsutum) was investigated at the biochemical level on the basis of aconitase (ACO), malate dehydrogenase (MDH), peroxidase (PER), phosphoglucoisomerase (PGI), phosphoglucomutase (PGM), and shikimate dehydrogenase (SKDH) isozyme banding patterns. Results revealed that five of the six isozymes are polymorphic among racial stocks of Upland cotton. Three of the six isozymes exhibit monomorphic banding patterns among the cultivars of Upland cotton.Taxonomic relationships among selected diploid and tetraploid species of Gossypium were evaluated on the basis of ACO, SKDH, PGM, MDH and PER isozyme systems. Multivariate analyses revealed that G. hirsutum and G. lanceolatum have a very close relationship. G. mustelinum was found to be very distinct from the other tetraploid species. A and D genome diploids were found to be most closely related to the tetraploid species, as expected based on previous cytogenetic evidence that these two genomes are the possible progenitors of the tetraploid species (AD genomes) of Gossypium. |
