Comparative physical mapping of the fibronectin and gamma crystallin genes in the mouse and cow, and karyotypic analysis of a panel of bovine-hamster hybrid somatic cells /

| Abstract: | The genes for fibronectin, gamma crystallin and isocitrate dehydrogenase are syntenic in man, mouse and cow. In an effort to physically localize this conserved chromosomal region in the genomes of the respective species, the genes for fibronectin and gamma crystallin have been localized to mouse chromosome 1, region C1-5, and to cow chromosome 8, region 1.1-1.4. Localizing these genes in both mouse and cow enables a cytological comparison of the conserved chromosomal region containing these genes in their respective species, and also enables comparisons of the genetic map with the physical map of mouse chromosome 1. Chromosomal identification in the mouse was achieved using a Robertsonian translocated marker chromosome, and in the cow, a cow-hamster hybrid somatic cell panel was utilized. Confirmation of chromosomal identity and regional localization was made using the technique of in situ hybridization to metaphase chromosomes. Further analysis of this hybrid somatic cell panel revealed tentative chromosomal assignments for two additional bovine syntenic groups, and confirmation of two groups previously localized to chromosomes. Tentatively assigning these additional syntenic groups to cow chromosomes increases the number of genes physically mapped to 9 autosomes and the X chromosome. Comparisons of synteny of homologous gene loci among man, mouse and cow support the generalization that a high degree of evolutionary conservation of chromosomal segments exists between human and mouse, with even more extensive conservation between human and cow. |
