520 3 |a The development of maize kernels cultured in vitro at three different temperatures was studied under controlled environmental conditions. Abscisic acid (ABA), soluble carbohydrates, soluble proteins, dry weight and fresh weight were assayed in both embryo and endosperm tissue at intervals from 11 to 35 days after pollination (DAP). Data were compared with those from field-grown kernels of comparable ages. Temperature effects caused modifications in the rate of kernei development and the grain filling period. High temperature accelerated development rate and reduced the grain filling period at least 5 days. At low temperature, development was slower and the grain filling period appeared to be longer than in the other temperature treatments. Embryo ABA apparently was not affected by different temperature treatments until 30 DAP, when ABA levels declined at the low temperature and increased drastically at the high temperature. Endosperm grown at 25/20 °C had high ABA levels after 15 DAP, apparently caused by low temperature. This suggests that ABA had a reduced turnover rate in endosperms grown at low temperature. If the high levels measured in endosperm grown at low temperature were due to reduced turnover rate, then the different temperature effects noted in embryo and endosperm suggest that ABA turnover may be differentially regulated in these tissues..
