| Abstract: | Several domesticated inbred lines of corn from the United States as well as open pollinated lines from Canada and Mexico were assayed for quantitative differences in amounts of chromosomal heterochromatin and genome size. A 16.9% difference in heterochromatin was observed among lines with Tama knobless flint, a Canadian line, having the lowest amount and Tx601, an inbred line, having the highest amount. Among the inbred lines NY302 (from New York) has the lowest heterochromatin amount (1%) and Tx601 (from Texas) the highest amount, 16.9%. Significant differences (up to 23%) in genome size (DNA content) were observed among lines. Gaspe flint had the smallest and Zapalote Grande the largest genome. Among the inbred lines, SD9 (a South Dakota line) had the smallest genome size and Tx601 had the largest. Genome size and amount of heterochromatin were found to be significantly correlated (r = .77). Fluorochrome staining after C-banding revealed that the heterochromatic areas fluoresce regardless of whether A-T or G-C enhancing fluorochromes were used. Two fluorochromes, DAPI and Hoechst, revealed differential fluorescence among types of corn heterochromatin. |
