| Abstract: | A reliable method for collecting semen from common marmosets via electroejaculation was perfected. Semen samples were obtained in approximately 95% of the attempts, and they contained viable, structurally intact spermatozoa as confirmed by light and electron microscopy. The procedure should be suitable in any situation for which it is necessary to obtain marmoset spermatozoa for study. In a morphologic study marmoset sperm populations were found to be structurally homogeneous with the majority of the spermatozoa concentrated in a viscous seminal coagulum. Percent motility was generally above 60% and most of the spermatozoa had good forward progression unless they were entrapped in the coagulum. Basically, the marmoset spermatozoon resembled that of other primates. Spermatozoa measured 50 (mu)m in total length. The head was dorsoventrally flattened and measured 3 (mu)m in width and 5 (mu)m in length. The middle piece contained 20-25 mitochondrial gyres and joined the principal piece without the distinct annulus reported in other mammals. Abnormal spermatozoa were observed in all samples examined, although less than 1% of the spermatozoa in any one sample were found to be defective. Anomalies of the head included invaginated acrosomes, double acrosomes, malformed acrosomes and invaginated nuclei. A single example of supernumary dense fibers was discovered in the middle piece of one spermatozoon, and several spermatozoa with disorganized or missing microtubules were encountered. Cytoplasmic droplets surrounding the middle piece were common findings although spermatozoa with droplets did not exhibit other signs of immaturity.
