520 3 |a In order to investigate changes in nucleotide metabolism during a developmental change in fungus Neurospora crassa, intracellular nucleotide pools were determined b high pressure liquid chromatography before and during conidial germination. Wild type, as well as mutants blocked in de novo purine biosynthesis were used to characterize metabolic fluxes in the purine, pyrimidine, and histidine biosynthetic pathways which are known to be interrelated in Neurospora. These studies showed that the purine and pyrimidine biosynthetic pathways were active prior to and during germination, even when exogenous adenine was used to satisfy the auxotrophic requirement for purines in the mutant strains. However, in germinated conidia starved for adenine, large increases in all non-adenine containing nucleotides were found, showing that purine and pyrimidine biosynthesis are stimulated by the absence of an end-product, and also suggesting competition for the common substrate 5-phosphoribosyl-1-pyrophosphate (PRPP) limits the over-all rate of purine and pyrimidine biosynthesis. ...
